When panelising, can all the panel area be used, or is some space needed for
handling etc ?
(ed) eg the 10cm square standard size is cheap , can it be split into 4 x 5x5
or 2 x 10x5 parts ?

You can split it into the sizes you state, but you are absolutely correct - you need to leave a little room around the edge for the V-grooving:
In the above image, note that the OUTLINE layer defines the four separate boards on the 100x100 panel, with the dividing lines being at 50mm horizontal and 50mm vertical.
The boards themselves though CANNOT be EXACTLY 50mm square, you need to design them to be at least 1mm narrower to allow room for the V-groove.
In the example above, each of those boards is actually 49mm x 49mm rather then 50 x 50 if you see what I am getting at.
Done like that, you can snap the boards out of the V-groove panel, and they will always be perfect.
Smoke makes things work. When the smoke gets out, it stops!
